Powder Injection
AlgoRx's core drug delivery technology involves the acceleration of dry, solid powder-formulations of medicines to high speed, using a transient high velocity helium gas jet, for injection into any physically accessible tissue. These particles must have suitable properties and fall in a specific size range. The particles may consist of pure medicine or advanced formulations containing additional inert ingredients to dilute or stabilize the product. The technology can be used to deliver traditional small molecules, peptides, or protein drugs. AlgoRx has identified inert ingredients that alter the physical properties of the particles to optimize them for delivery. The basic principle underlying this technology is that suitable particles can be reproducibly delivered into the body with high efficiency if they are appropriately formulated and are traveling at a sufficiently high velocity to breach the outermost skin barrier, the stratum corneum. Thus the technology can be applied to any medicine that can be formulated into solid particles of the appropriate size distribution, mass (i.e. density) and strength.
The Powder Injection System is the combination of a delivery device with a specially formulated powdered medicine. Separate devices have been configured for injection into normal skin and into mucosal sites. Some devices are designed for single use and are completely disposable and others, intended for longer courses of therapy, have reusable elements. The principle behind the operation of the devices is the same: harnessing the energy of a transient gas jet to accelerate a pre-measured dose of powder formulation particles so that they may superficially enter the chosen tissue. As many elements of AlgoRx's delivery devices are common, a small number of device configurations will provide many applications.

Powder Injection Systems are powered by a custom-manufactured helium gas micro cylinder and use a cassette or package to introduce the powder-form medicine into the gas flow. The gas micro cylinder is deep-drawn from aluminum and is filled with helium to a specified pressure and laser-welded. To operate, the device is applied to the tissue site to be treated, and an actuation pin breaks the gas cylinder tip. This releases the compressed gas, which bursts the drug cassette and delivers the drug to the tissue. For all design configurations and applications the rest of the device is manufactured from medical-grade plastics using standard injection-molding techniques.
For accurate, high-throughput automated manufacture, 2-3 mg per dose is currently the most desirable payload range. For higher payloads delivery efficiency must be optimized. Different device design configurations are available depending on the specific application intended.
